
Pipetting Master Mixes and Samples
New generation diagnostic kits are using 1-Step RT-PCR master mixes which includes the cDNA enzime to be able to complete the PCR protocol as a single RUN, without any additional steps, without any interruption.
Almost all kits has a single master mix tubes, PC and NTC inside.
Firstly, master mix is pipetted into all tubes, because no amplification plots could be obtained unless RT-PCR enzymes (cDNA and PCR) are used.
- RT-PCR Mix = Reaction Mix (Needs for PCR reaction)
Secondly, one of the 3 liquid must be pipetted into each single tube;
- PC = Positif Control (To check the instrument and kit is working properly)
- NTC = Non Template Control (To check whether there is a contamination into mixes or not)
- Sample = Patient (RNA Prep solution that you put the swap inside)
About PCR 8 Strip Tubes
To work with high efficiency 0.1 ml white (not clear) PCR strip tubes and 0.1 ml white (not clear) 96 well plates must be used.
